The short answer is as soon as there is a 2nd degree exemplification offered.  There is no "waiting period" or things to accomplish before a 1st degree Brother can advance.  We encourage you to move to full knighthood (attainment of the 3rd degree) as quickly as you can so that you may fully appreicate all that being a Knight of Columbus means.
